
Purifying partials for use as
reference material
Purifying partially filled AAV9 capsids
We were asked to purify partial capsid species for an AAV9 construct packaging a particular transgene of interest. To do this, we ran our upstream and downstream optimisation routines in parallel, seeking conditions which both generated partials and allowed us to separate fractions containing partials of differing sizes.
This material can be used to support analytical method development such as spike-ins for interpreting viral genome sequencing data and as controls for ddPCR, AUC, or CD-MS.

Following successful purification with AAV9, we then sought to determine if the same approach could be used to purify out partials from an engineered serotype. By tuning upstream and downstream and making use of a wide set of previously optimised conditions, we can isolate empty, full and partial capsids and formulate these for analytical development teams.
Analytical method development can be rapidly accelerated with the availability of a suite of construct specific reference samples. These can also be used for advanced safety and potency studies, as well as stability studies ahead of formulation.
