              Custom process                     development ahead of large          batch manufacturing            

Optimisation enables scalable manufacturing

We run high throughput upstream optimisation before every large batch. The scope is dependent on developer goals so that we can meet different product attributes.

We read out both yield (via dPCR), and quality (via mass photometry) for each process condition and process step.

Completed NHP-grade batches​​​​

Displayed are three large manufacturing batches (1-4e14 vg). After optimization, we obtain a high proportion of full capsids after purification using chromatography (Fig 2).

 

Final mass photometry data for a large-batch AAV9 project and associated CoA is displayed in the top two panels.  The bottom two panels show mass photometry plots for two other large-batch projects: an undisclosed known serotype (left), and undisclosed engineered capsid that was refractory to methods used to purify the serotype it derived from (right).
