 Rapid optimisation workflows to try multiple capsid variants 

Rapid optimisation workflows enable us to test one transgene in many serotypes

We conducted a feasibility study to test a single transgene in multiple capsids.  Rapid optimisation workflows enable us to develop bespoke conditions for capsid and transgene pairs (Fig 1).
 

These conditions enable discovery scientists to make quick assessments of optimal capsid choice with a single transgene design in early discovery work, while knowing they can progress rapidly to NHP studies as more data readouts come in.

We optimise early on in case any candidates progress to large scale manufacturing​​​​

By onboarding constructs early in the development cycle, we can rapidly progress to large batch manufacturing whenever it is needed.

 

After screening for yield, we often choose to tune the upstream conditions to modulate quality.  This is an example of a second optimisation round which proved valuable when later-stage manufacturing was required.
